it depends what you mean, a continuous tradition keeping near the exact same curriculum as when it was founded? A cultural practice that is continuous but changes with each generation, maybe not even with a fixed curriculum? Do reconstructions count?

And then I would simply defer to the oldest culture. So if Homo Sapiens originated in east Africa, then I would suggest maybe Nuba wrestling? But the thought occurred to me that perhaps there were martial arts before Homo Sapiens...

It's claims to historicity mostly rely on chinese apochrypal texts from the 17th century claiming to be about a man who supposedly lived in the 5~6th century. In all likelihood kalaripayattu as it exists today is only a couple hundred years old at best.
